Recoding the pioneer factor Ascl1 to reveal and rewire the molecular logic of neuronal reprogramming
Ascl1 is a transcription factor that can reprogram fibroblasts into neurons, but the molecular mechanisms remain poorly understood. We will use advanced single-molecule techniques to map how Ascl1鈥檚 sequence and structure control its function on chromatin. This mechanistic insight will enable the design of optimized Ascl1 variants, paving the way for more efficient neuronal reprogramming and future regenerative therapies.
